New drug targets revealed from giant parasitic worm genome sequence

Scientists have identified the genetic blueprint of the giant intestinal roundworm, Ascaris suum, revealing potential targets to control the devastating parasitic disease, ascariasis which affects more than one billion people in China, South East Asia, South America and parts of Africa, killing thousands of people annually and causing chronic effects in young children.

Dr Aaron Jex and Professor Robin Gasser from the University of Melbourne's Faculty of Veterinary Science led the international research project which was published in the journal Nature today [Oct. 27].

The work has provided new insights to treat the condition ascariasis, identified by the World Health Organization as a key, neglected disease in urgent need of extensive research and significantly improved control.

"Sequencing the genome of Ascaris suum is a major step towards controlling the infection it causes because the more we know about the genetics of the parasite and how it works, the better we can fight it with novel treatments," Dr Jex said.

"From the genome sequence we have identified five high priority drug targets that are likely to be relevant for many other parasitic worms. New treatments are urgently needed and genome-guided drug target discovery is ideal for identifying targets that selectively kill the parasite and not the host.

"We also identified key information on how the parasite hides from the immune system, which is essential for any future vaccine development."

The team sequenced the nuclear genome of Ascaris suum which is an easier worm to study in the laboratory and mainly only infects pigs, but is very closely related to Ascaris lumbricoides which causes ascariasis in humans. The worms range from 15-30cm in length and are spread via eggs in contaminated food.

Ascaris lumbricoides is one of the most common parasites of humans, affecting more than one billion people in developing countries, particularly children, causing impaired physical and cognitive development, and in severe cases death, due to lack of nutrient absorption and intestinal blockage.

Ascaris suum also causes major production losses in swine farming due to reduced growth, failure to thrive and mortality.

GOP senators vote to defeat Obama's jobs bill (AP)

WASHINGTON ? United against Barack Obama, Senate Republicans voted Tuesday night to kill the jobs package the president had spent weeks campaigning for across the country, a stinging loss at the hands of lawmakers opposed to stimulus-style spending and a tax increase on the very wealthy.

The $447 billion plan died on a 50-49 tally that garnered a majority of the 100-member Senate but fell well short of the 60 votes needed to keep the bill alive. The tally had been 51-48, but Senate Majority Leader Harry Reid, D-Nev., switched his vote to "nay" so that he could force a future revote.

The demise of Obama's jobs package was expected, despite his campaign-style efforts to swing the public behind it. The White House and leaders in Congress were already moving on to alternative ways to address the nation's painful 9.1 percent unemployment, including breaking the legislation into smaller, more digestible pieces and approving long-stalled trade bills.

"Tonight's vote is by no means the end of this fight," Obama said in a statement after the vote. "Because with so many Americans out of work and so many families struggling, we can't take `no' for an answer."

The White House appears most confident that it will be able to continue a 2-percentage-point Social Security payroll tax cut through 2012 and to extend emergency unemployment benefits to millions of people ? if only because, in the White House view, Republicans won't want to accept the political harm of letting those provisions expire.

White House officials are also hopeful of ultimately garnering votes for the approval of infrastructure spending and tax credits for businesses that hire unemployed veterans.

"Now it's time for both parties to work together and find common ground on removing government barriers to private-sector job growth," House Speaker John Boehner, R-Ohio, said after the vote.

Democrats Ben Nelson of Nebraska and Jon Tester of Montana ? both up for re-election next year in states where Obama figures to lose ? broke with their party on Tuesday night's vote. Every Republican present opposed the plan.

Earlier in the day, Obama capped his weekslong campaign for the measure in an appearance typical of the effort ? a tough-talking speech in a swing state crucial to his re-election. Like earlier appearances, it seemed aimed more at rallying his core political supporters heading into the election than changing minds on Capitol Hill.

"Any senator who votes no should have to look you in the eye and tell you what exactly they're opposed to," Obama said to a union audience in Pittsburgh. "I think they'll have a hard time explaining why they voted no on this bill ? other than the fact that I proposed it."

Democrats were not wholly united behind the measure. In addition to Nelson and Tester, Sens. Jim Webb, D-Va., Joe Manchin, D-W.Va., and Joe Lieberman, a Connecticut independent who aligns with Democrats, said they oppose the underlying measure despite voting to choke off the filibuster.

Obama's plan would combine Social Security payroll tax cuts for workers and businesses and other tax relief totaling about $270 billion with $175 billion in new spending on roads, school repairs and other infrastructure, as well as unemployment assistance and help to local governments to avoid layoffs of teachers, firefighters and police officers.

Obama said that the plan ? more than half the size of his 2009 economic stimulus measure ? would be an insurance policy against a double-dip recession and that continued economic intervention was essential given slower-than-hoped job growth.

"Right now, our economy needs a jolt," Obama said. "Right now."

Unlike the 2009 legislation, the current plan would be paid for with a 5.6 percent surcharge on income exceeding $1 million. That would be expected to raise about $450 billion over the coming decade.

"Democrats' sole proposal is to keep doing what hasn't worked ? along with a massive tax hike that we know won't create jobs," Senate Minority Leader Mitch McConnell, R-Ky., said Tuesday, saying there are 1.5 million fewer jobs than when Obama's 2009 economic package became law. "Why on earth would you support an approach that we already know won't work?" McConnell said.

The White House and Democratic leaders, however, were pleased that the great majority of Democrats voted for the plan. Support among Democrats was shored up by replacing Obama's tax increases ? particularly a proposal to limit the value of itemized deductions for families making more than $250,000 ? with the surcharge on annual income over $1 million.

That millionaires proposal would hit about 392,000 households, according to an analysis by the Tax Policy Center, a Washington think tank. In 2013, the first year the tax would take effect, those wealthy households would see their taxes increase by an average of $110,500, according to the analysis.

Just before the vote on Obama's jobs plan, the Senate passed legislation aimed at punishing China for keeping its currency undervalued against the dollar. Lower-valued currency helps Chinese exports at the expense, bill supporters say, of American jobs.

Next, both the House and Senate will turn Wednesday to approving trade agreements with Colombia, Panama and South Korea that could create tens of thousands of jobs, one of the few areas of agreement between Republicans and the administration on boosting the economy.

In coming weeks and months, Democrats promise further votes on jobs. But it remains to be seen how much of that effort will involve more campaign-stoked battles with Republicans and how much will include seeking common ground in hopes of passing legislation.

Leaders of the GOP-controlled House have signaled they support tax cuts for small businesses and changes to jobless insurance to allow states to use unemployment funds for on-the-job training. And they've indicated they'll be willing to accept an extension of cuts to the Social Security payroll tax. But stimulus-style spending is a nonstarter with the tea party-infused chamber.

Tuesday's vote played out as disaffected crowds continued to occupy Wall Street, a square in Washington and parts of other cities around the country in protest of income inequality and related issues.

Obama advisers said they were working with Senate Democratic leaders on how and when to break out separate aspects of the overall jobs bill for votes.

Even before Tuesday's defeat, the White House was casting the Senate vote as but the first act in what one Obama adviser called a long-term play ? essentially, an autumn full of action to force Congress to take action on jobs. Senior Obama officials said it was important for the Senate to act to keep pressure on the Republican-led House.

Sen. Tom Coburn, R-Okla., was recuperating from surgery and did not vote.

'Dark matter' of the genome revealed through analysis of 29 mammals

ScienceDaily (Oct. 12, 2011) ? An international team of researchers has discovered the vast majority of the so-called "dark matter" in the human genome, by means of a sweeping comparison of 29 mammalian genomes. The team, led by scientists from the Broad Institute, has pinpointed the parts of the human genome that control when and where genes are turned on. This map is a critical step in interpreting the thousands of genetic changes that have been linked to human disease.

Their findings appear online October 12 in the journal Nature.

Early comparison studies of the human and mouse genomes led to the surprising discovery that the regulatory information that controls genes dwarfs the information in the genes themselves. But, these studies were indirect: they could infer the existence of these regulatory sequences, but could find only a small fraction of them. These mysterious sequences have been referred to as the dark matter of the genome, analogous to the unseen matter and energy that make up most of the universe.

This new study enlisted a menagerie of mammals -- including rabbit, bat, elephant, and more -- to reveal these mysterious genomic elements.

Over the last five years, the Broad Institute, the Genome Institute at Washington University, and the Baylor College of Medicine Human Genome Sequencing Center have sequenced the genomes of 29 placental mammals. The research team compared all of these genomes, 20 of which are first reported in this paper, looking for regions that remained largely unchanged across species.

"With just a few species, we didn't have the power to pinpoint individual regions of regulatory control," said Manolis Kellis, last author of the study and associate professor of computer science at MIT. "This new map reveals almost 3 million previously undetectable elements in non-coding regions that have been carefully preserved across all mammals, and whose disruptions appear to be associated with human disease."

These findings could yield a deeper understanding of disease-focused studies, which look for genetic variants closely tied to disease.

"Most of the genetic variants associated with common diseases occur in non-protein coding regions of the genome. In these regions, it is often difficult to find the causal mutation," said first author Kerstin Lindblad-Toh, scientific director of vertebrate genome biology at the Broad and a professor in comparative genomics at Uppsala University, Sweden. "This catalog will make it easier to decipher the function of disease-related variation in the human genome."

This new map helps pinpoint those mutations that are likely responsible for disease, as they have been preserved across millions of years of evolution, but are commonly disrupted in individuals that suffer from a given disease. Knowing the causal mutations and their likely functions can then help uncover the underlying disease mechanisms and reveal potential drug targets.

The scientists were able to suggest possible functions for more than half of the 360 million DNA letters contained in the conserved elements, revealing the hidden meaning behind the As, Cs, Ts, and Gs. These revealed:

  • Almost 4,000 previously undetected exons, or segments of DNA that code for protein
  • 10,000 highly conserved elements that may be involved in how proteins are made
  • More than 1,000 new families of RNA secondary structures with diverse roles in gene regulation
  • 2.7 million predicted targets of transcription factors, proteins that control gene expression

"We can use this treasure trove of new elements to revisit disease association studies, focusing on those that disrupt conserved elements and trying to discern their likely functions," said Kellis. "Using a single genome, the language of DNA seems cryptic. When studied through the lens of evolution, words light up and gain meaning."

The researchers were also able to harness this collection of genomes to look back in time, across more than 100 million years of evolution, to uncover the fundamental changes that shaped mammalian adaptation to different environments and lifestyles. The researchers revealed specific proteins under rapid evolution, including some related to the immune system, taste perception, and cell division. They also uncovered hundreds of protein domains within genes that are evolving rapidly, some of which are related to bone remodeling and retinal functions.

"The comparison of mammalian genomes reveals the regulatory controls that are common across all mammals," said Eric Lander, director of the Broad Institute and the third corresponding author of the paper. "These evolutionary innovations were devised more than 100 million years ago and are still at work in the human population today."

In addition to finding the DNA controls that are common across all mammals, the comparison highlighted areas that have been changing rapidly only in the human and primate genomes. Researchers had previously uncovered two hundred of these regions, some of which are linked to brain and limb development. The expanded list -- which now includes more than 1,000 regions -- will give scientists new starting points for understanding human evolution.

The comparison of many complete genomes is beginning to offer a clear view of once indiscernible genomic regions, and with additional genomes, that resolution will only increase. "The power of this resource is that it continues to improve with the inclusion of more species," said Lindblad-Toh. "It's a very systematic and unbiased approach that will only become more powerful with the inclusion of additional genomes."

John Wayne auction fetches more than $5 million (AP)

LOS ANGELES ? Organizers say a two-day auction of more than 700 costumes, scripts, awards and memorabilia from the estate of film star John Wayne brought in $5.4 million.

Heritage Auctions said Friday that the auction included items such as Wayne's last driver's license, a holster and gun belt from the film "El Dorado" and an eye patch from the 1969 movie, "True Grit."

The green wool beret Wayne wore in "The Green Berets," went for $179,250 and a cowboy hat worn in "Big Jake" and "The Cowboys" fetched $119,500.

The items had been kept by Wayne's family since his 1979 death from cancer.

A portion of the proceeds will be used by the John Wayne Cancer Foundation to help fund cancer research, treatment and education.

3. what is meant by relatives will interfere and examine the relationship and not let us be. we would be defending the relationship rather than living it and having to convince the family.

he panicked when that happened and the next day that was when it was over between us. he has told me since that her happiness comes before his and no matter what he cant lose her. so i know that his mother was the main reason for us breaking up. i dont understand how you can make a son choose when he is so happy. he pleaded with her and told her he couldnt live without me, but i also know what if he wanted me so much he would have left. in my heart i know he was a coward and not strong enough and he walked away and left me, but im devastated cos ive lost the best person i ever knew. i know i wont find a love like this again, what i felt for him will be hard to replicate with someone else. at my age also, i feel that this might have been my last chance at happiness and maybe now i should also give up too on love and accept being on my own. im 43 he was 26.

yes we did meet..we had a couple of holidays each lasting two weeks and we were inseparable. he doesnt want children at all..and i mean he is seriously not having any. even if i was younger and i got pregnant he would have gone crazy cos he doesnt want to be a father. he is totally against the idea cos of illness in his family chain. it wasnt online only. it was real too. we were planning a future and a life here in the uk with me. we were very happy but choosing between me and his parents made him scared. he said he has torn himself apart over this until he reached the conclusion that it was either me or the people who raised him. i can still have kids..i havent had any yet and im fit and healthy. women here in the uk have kids till 48 easily. his own mother had him at 37. shes the same age as my mother..ie in her 60?s. so is his father, as its second marriage.

Different religious ideas. I have a life independent of his family and they don?t like it. I tell him numerous times to hang out with them and he does every now and then. Moody's cuts ratings on Intesa, UniCredit (Reuters)

MILAN (Reuters) ? Moody's lowered its credit ratings on Italy's two biggest banks, Intesa Sanpaolo (ISP.MI) and UniCredit (CRDI.MI), on Wednesday following its recent downgrade of Italian sovereign debt.

On Tuesday, Moody's lowered its rating on Italy's bonds by three notches to A2, saying it saw a "material increase" in funding risks for euro zone countries with high levels of debt and warning that further downgrades were possible.

Italian banks have come under pressure in recent weeks because of the spreading debt crisis and are seen as vulnerable because of their vast holdings in Italian government bonds.

Rising government bond yields have also driven funding costs higher for domestic lenders.

Moody's said in a note it had cut UniCredit's long-term debt and deposit ratings to A2 from Aa3, concluding the review for possible downgrade initiated in May this year.

"Growth prospects for many of the group's key markets, and in particular Italy, have been cut in recent months, which is likely to make significant further improvement in profitability and asset quality more uncertain and difficult to achieve," Moody's said.

But the rating agency also pointed up the bank's improved capital adequacy and its robust liquidity, which has allowed it to cope with difficult market funding conditions.

The long-term debt of Intesa, Italy's biggest retail bank, was cut to A2 from Aa3. In a note Moody's noted Intesa's direct exposure to the Italian sovereign and its pronounced focus on domestic business.

The management of both UniCredit and Intesa Sanpaolo have said they currently have no funding problems.

Besides its downgrade of Italian banks, Moody's also cut its ratings on the debt of a series of state-controlled Italian companies including oil and gas group Eni (ENI.MI), utility Enel (ENEI.MI), defense company Finmeccanica (SIFI.MI) and power grid operator Terna (TRN.MI).

ING Group?s European real estate investment management operations, ING REIM Europe, will be brought into the fold before the end of the year; CBRE Group anticipates a closing during the third quarter. When all is said and done, the firm will have relied on $400 million of new term loans and cash on hand to finance the REIM Asia and REIM Europe purchases. CBRE Group had raised $800 million of bank debt for the three-phase ING transaction, and spent the other $400 million to finance the first phase of the three-part deal, the acquisition of ING Clarion Real Estate Securities, which closed at the end of the second quarter.

First comet found with ocean-like water

New evidence supports the theory that comets delivered a significant portion of Earth's oceans, which scientists believe formed about 8 million years after the planet itself.

The findings, which involve a University of Michigan astronomer, are published Oct. 5 online in Nature.

"Life would not exist on Earth without liquid water, and so the questions of how and when the oceans got here is a fundamental one," said U-M astronomy professor Ted Bergin, "It's a big puzzle and these new findings are an important piece."

Bergin is a co-investigator on HiFi, the Heterodyne Instrument for the Infrared on the Hershel Space Observatory. With measurements from HiFi, the researchers found that the ice on a comet called Hartley 2 has the same chemical composition as our oceans. Both have similar D/H ratios. The D/H ratio is the proportion of deuterium, or heavy hydrogen, in the water. A deuterium atom is a hydrogen with an extra neutron in its nucleus.

This was the first time ocean-like water was detected in a comet.

"We were all surprised," Bergin said.

Six other comets HiFi measured in recent years had a much different D/H ratio than our oceans, meaning similar comets could not have been responsible for more than 10 percent of Earth's water.

The astronomers hypothesize that Hartley 2 was born in a different part of the solar system than the other six. Hartley most likely formed in the Kuiper belt, which starts near Pluto at about 30 times farther from the sun than the Earth is. The other six hail from the Oort Cloud more than 5,000 times farther out.

The source of earth's oceans has been a subject for debate among astronomers for decades. Until now, asteroids were thought to have provided most of the water. Now, however, Herschel has shown that at least one comet does have ocean-like water.

"The results show that the amount of material out there that could have contributed to Earth's oceans is perhaps larger than we thought," Bergin said.

J.R. Martinez on Dancing With the Stars: An Inspiring, Tear-Jerking Performance


We didn't know it was possible to become legitimately choked up while watching Dancing With the Stars, but J.R. Martinez proved it possible Monday night.

There weren't many dry eyes left in the room following the Iraq War veteran's moving life story of injury and redemption, followed by an emotional dance.

After J.R. finished his rumba with Karina Smirnoff, the All My Children actor seemed taken aback by what he's been through - not just on DWTS but in life.
